Issue Date: | 2014 | Abstract: | MS is the most common neurological disease in young adulthood. Although current treatments have achieved some progress in the treatment of RR MS patients, these therapies are not effective in the secondary progressive phase of the disease. Likewise, no agent has been shown to affect primary progressive MS. The pathophysiology of MS is heterogeneous, and it has been suggested that different pathological subtypes of MS may require different therapeutical approaches. In this study, first we developed and validated a murine IL-11 encoding lentiviral vector. Secondly, by using a mouse model that mimics the pattern III MS lesion subtype, we explored the therapeutic effect of IL-11 on deand remyelination processes. Additionally, we investigated the effect of recombinant murine IL-11 on myelin phagocytosis and on the M1/M2 phenotype of microglial cells. MS is de meest voorkomende neurologische ziekte bij jongvolwassenen. Hoewel de huidige medicatie enige vooruitgang geboekt heeft bij de behandeling van relapsing remitting (RR) MS- patiƫnten, zijn ze weinig effectief in de secundair progressieve fase van de ziekte. Bovendien is er geen goed geneesmiddel voor primair progressieve MS. De pathofysiologie van MS is heterogeen, wat suggereert dat verschillende pathologische subtypes van MS telkend een andere therapeutische benadering nodig hebben. In deze studie, werd eerst een recombinant muis IL-11 coderende lentivirale vector (LV) ontwikkeld en gevalideerd. Vervolgens werd een muismodel dat het MS-type III lesie nabootst gebruikt om het therapeutisch effect van IL-11 op de- en remyelinisatie processen te onderzoeken. Tot slot werd het effect van recombinant muis IL-11 onderzocht op myeline fagocytose en het induceren van een M1 of M2 fenotype in microgliale cellen. |
