dc.description.abstractInplanarians,pluripotencycanbestudiedinvivointheadultanimal,makingtheseanimalsauniquemodelsystemwherepluripotency-basedregeneration(PBR)—anditstherapeuticpotential—canbeinves-tigated.Thisreviewfocusesonrecentfindingstobuildacloudmodeloffaterestrictionlikelihoodforplanarianstemandprogenitorcells.Recently,acomputationalapproachbasedonfunctionalandmolec-ularprofilingatthesinglecelllevelwasproposedforhumanhematopoieticstemcells.Basedondatageneratedbothinvivoandexvivo,wehypothesizedthatplanarianstemcellscouldacquiremultipledirec-tionlineagebiases,followinga“badlands”landscape.Insteadofadiscretetree-likehierarchy,wherethepotencyofstem/progenitorcellsreducesstepwise,weproposeaContinuumofLOw-primedUnDiffer-entiatedPlanarianStem/ProgenitorCells(CLOUD-PSPCs).Everysubclassofneoblast/progenitorcellsisacloudoflikelihood,asthesinglecelltranscriptomicsdataindicate.TheCLOUD-HSPCsconceptwassubstantiatedbyinvitrodatafromcellculture;therefore,toconfirmtheCLOUD-PSPCsmodel,thepla-nariancommunityneedstodevelopnewtools,likelivecelltracking.FuturestudieswillallowadeeperunderstandingofPBRinplanarian,andthepossibleimplicationsforregenerativetherapiesinhuman.-
