Issue Date: | 2022 | Abstract: | Soil pollution by the metal cadmium (Cd) mainly originates from anthropogenic
activities and is a worldwide and severe problem. Even though Cd is a toxic, nonessential metal, it is readily taken up by plants and introduced into the food chain,
posing risk to human health. Once inside the plant, Cd disturbs various
developmental and physiological processes, compromising plant productivity.
From a cellular perspective, Cd toxicity mainly arises from its ability to, indirectly,
stimulate the production of reactive oxygen species (ROS), resulting in an
oxidative challenge. Even though ROS are major damaging agents, they also fulfil
important signalling roles. Hydrogen peroxide (H2O2), for example, is considered
a prominent signalling molecule. Nevertheless, to allow ROS signalling and, at the
same time, prevent oxidative damage, ROS levels must be tightly balanced and
excessive ROS production must be prevented. To do so, plants rely on a wide
range of enzymatic and non-enzymatic antioxidants. One of the most abundant
and important antioxidants is the metabolite glutathione (GSH). Glutathione
serves to detoxify ROS, like H2O2, directly but mainly through the ascorbate (AsA)-
GSH cycle. Besides its function as antioxidant, the presence of a thiol group
enables GSH and its oligomers, termed phytochelatins (PCs), to chelate Cd and
sequester it into the vacuole. Previous research demonstrated that Cd exposure
results in the rapid depletion of root GSH levels due to its allocation to PC
synthesis. This impacts the antioxidative capacity of GSH in the early responses
to Cd stress. Moreover, the depletion of this ubiquitous antioxidant will most likely
trigger specific signalling events that define the acute responses and acclimation
to Cd stress. Besides alterations in the GSH pool and H2O2 signalling, the
phytohormone ethylene functions as important regulator in the responses to Cd
stress. Ethylene was previously implemented as mediator of the early Cd-induced
oxidative challenge in Arabidopsis thaliana based on the inability of the ethylene
insensitive 2-1 (ein2-1) mutant to stimulate leaf GSH levels upon Cd exposure. In
addition, it was suggested that ethylene signalling is involved in the stimulation
of ROS production in response to Cd through NADPH oxidases. To increase the spatiotemporal resolution, a time-course analysis was conducted
in order to unravel the sequence of events in terms of short-term Cd-induced
responses focussing on the key regulators described above. The use of a shortterm exposure set-up (0 h to 24 h) allowed for the identification of pressure points
prior to acclimation. Responses related to GSH, 1-aminocyclopropane-1-
carboxylic acid (ACC), ethylene, H2O2 and the oxidative challenge were studied at
the metabolite and/or transcript level in roots and leaves of A. thaliana exposed
to 5 µM Cd. Based on our findings, we propose that the early root GSH depletion
is required for a proper Cd-induced signalling response. Furthermore, we
demonstrated that root ACC levels are stimulated early on during the Cd-induced
stress response and that H2O2 levels are modulated by the overall GSH pool rather
than its redox state.
To obtain a better understanding of the importance of GSH during the short-term
Cd-induced stress response and elucidate how this is defined by the rapid GSH
depletion, Cd-induced root and leaf responses (described above) were compared
between wild-type (WT) A. thaliana plants and GSH-deficient cadmium-sensitive
2-1 (cad2-1) mutant plants. We demonstrated that even though the cad2-1
mutant has decreased GSH levels, root GSH depletion still occurs and the chelating
capacity of GSH is prioritised over its antioxidative function. Our study indicated
that under GSH-limiting conditions, the Cd-induced stimulation of GSH
metabolism and ACC production is accelerated, which is conceivably mediated by
a more pronounced oxidative challenge. Additionally, the redox imbalance in
cad2-1 mutant plants and the lack of proper transient ethylene signalling results
in suboptimal stabilisation and, consequently, a more pronounced Cd effect.
As previously indicated, ethylene signalling is important for the stimulation of leaf
GSH metabolism under Cd exposure and is involved in the Cd-induced oxidative
challenge. In this study, we aimed to unravel the involvement of ethylene
signalling in short-term Cd-induced root and leaf responses and assess how these
were altered by impaired ethylene signalling. By comparing the same responses
as described above between WT and ein2-1 mutant plants, we found that the role
of ethylene signalling in responses to Cd stress is largely similar in both organs.
More precisely, we demonstrated that in both organs the timing and the transient nature of the ethylene response is altered by impaired ethylene signalling and this
results in a delayed oxidative burst and lack of GSH stimulation under Cd
exposure. This untimely stress response affects stabilisation and possibly
acclimation.
As ACC biosynthesis was enhanced upon Cd exposure and stimulated by GSH
deficiency, we aimed to further elucidate the importance of ACC biosynthesis
during the short-term Cd-induced stress response. This time, root and leaf
responses were compared between WT A. thaliana plants and acs2-1acs6-1
double knockout mutant plants as both ACC synthase isozymes, ACS2 and ACS6,
were previously shown to be mainly responsible for the Cd-induced ethylene
biosynthesis. We showed that the impact of impaired ACC production was limited
and confined to 24 h, as became apparent by the delay in Cd-induced GSH
stimulation. While in WT plants the oxidative burst is typically established after 24
h of exposure and functions as an important signalling response, in the ACC
biosynthesis mutant this oxidative burst was delayed at the leaf level and not
observed in roots. Hence, we concluded that the Cd-induced oxidative challenge
is better controlled in WT plants compared to mutant plants as the latter suffer
from a delayed GSH stimulation and an untimely oxidative burst that is required
for proper signalling.
Taken together, this study provided a detailed view on the time course of Cdinduced stress responses and signalling events that precede acclimation. We
proposed that root GSH depletion is required for proper Cd-induced stress
signalling and showed that GSH deficiency accelerates the short-term stress
response which results in suboptimal acclimation. In addition, we showed that
impaired ethylene signalling and biosynthesis hinder Cd-induced GSH stimulation
in both roots and leaves and delay or impair the oxidative burst leading to an
untimely signalling response and a suboptimal or at least delayed acclimation. Bodemverontreiniging door het metaal cadmium (Cd) wordt voornamelijk veroorzaakt door menselijke activiteiten en vormt wereldwijd een ernstig probleem. Ondanks het feit dat Cd toxisch en niet-essentieel is, wordt het makkelijk opgenomen door planten en via deze weg geïntroduceerd in de voedselketen, wat een risico vormt voor de volksgezondheid. Eens door de plant opgenomen, verstoort Cd verschillende ontwikkelingsprocessen en fysiologische processen en wordt de plantengroei in het gedrang gebracht. Op cellulair niveau, wordt Cd-toxiciteit voornamelijk veroorzaakt door de indirecte productie van reactieve zuurstofsoorten (reactive oxygen species, ROS), wat leidt tot oxidatieve stress. Hoewel ROS schadelijk kunnen zijn, vervullen ze ook een belangrijke rol in signalering. Zo is waterstofperoxide (H2O2) een prominent signaalmolecule. Om ROS-signalering mogelijk te maken en tegelijkertijd schade te vermijden, moeten de ROS-niveaus binnen bepaalde limieten gehouden worden. Om dit te bewerkstelligen, berusten planten op een breed scala aan enzymatische en nietenzymatische antioxidanten. Een alomtegenwoordig en belangrijk antioxidant is het metaboliet glutathion (GSH). Glutathion speelt een belangrijke rol in de eliminatie van ROS, zoals H2O2. Dit kan rechtstreeks of via de ascorbaat (AsA)- GSH-cyclus. Naast zijn functie als antioxidant, zorgt de aanwezigheid van een thiolgroep in zijn structuur ervoor dat GSH en afgeleide oligomeren, fytochelatines (PCs) genaamd, Cd kunnen cheleren en sekwestreren in de vacuole. Voorgaand onderzoek heeft aangetoond dat Cd-blootstelling de snelle depletie van GSHniveaus in de wortel veroorzaakt doordat het gebruikt wordt voor de synthese van deze PCs. Dit vermindert de antioxidatieve capaciteit van GSH tijdens de vroege responsen onder Cd-stress. Bovendien zal de depletie van dit antioxidant verschillende signalen uitlokken die de acute responsen en acclimatie onder Cdstress definiëren. Naast de wijzigingen in GSH-niveaus en H2O2-signalering spelen plantenhormonen, zoals ethyleen, een belangrijke regulerende rol in Cdgeïnduceerde stressresponsen. Ethyleen werd voorheen reeds voorgesteld als een belangrijke regulator van Cd-geïnduceerde oxidatieve stress in Arabidopsis thaliana omwille van het feit dat de ethylene insensitive 2-1 (ein2-1) mutant niet in staat was de GSH-niveaus in bladeren te verhogen na Cd-blootstelling. Daarenboven werd gesuggereerd dat ethyleensignalering betrokken is bij de stimulatie van ROS-productie onder Cd-blootstelling via NADPH-oxidasen. Om de spatiotemporele resolutie van de verschillende regulatoren te vergroten, werd het tijdsverloop van verschillende acute Cd-geïnduceerde responsen geanalyseerd. Het gebruik van een korte termijn set-up (0 u tot 24 u), maakte het mogelijk om de drukpunten die voorafgaan aan acclimatie te identificeren. De responsen betreffende GSH, 1-aminocyclopropaan-1-carboxylzuur (ACC), ethyleen, H2O2 en oxidatieve stress en signalering werden bestudeerd op metaboliet- en/of transcriptniveau in zowel wortels als blaadjes van A. thaliana blootgesteld aan 5 µM Cd. Naar aanleiding van onze bevindingen wordt de Cdgeïnduceerde GSH-depletie op wortelniveau voorgesteld als een vereiste voor het induceren van een passende alarmrespons en het correcte verloop van de signalisatie tijdens de korte-termijn respons. Bovendien, werd aangetoond dat de ACC-concentraties gestimuleerd worden in het begin van de Cd-geïnduceerde stressrespons en dat de H2O2-niveaus eerder gemoduleerd worden door de totale GSH-concentratie dan door de redoxstatus van GSH. Om het belang van GSH tijdens de acute Cd-geïnduceerde stressrespons beter te begrijpen en om te verduidelijken in welke mate de GSH-depletie bepalend is voor deze responsen, werden de responsen vergeleken tussen wild-type (WT) A. thaliana planten en de GSH-deficiënte cadmium-sensitive 2-1 (cad2-1) mutant. Onze data toonden aan dat, ondanks de verminderde GSH-concentraties in de cad2-1 mutant, de GSH depletie in de wortels nog steeds optreedt en dat de chelerende eigenschappen van GSH prioriteit krijgen over de antioxidatieve functie. Daarenboven werd een vroegtijdige stimulatie in het GSH-metabolisme en de ACC-productie waargenomen. Dit is waarschijnlijk het gevolg van een meer uitgesproken oxidatieve stress die werd vastgesteld in de cad2-1 mutant. Daarbij resulteert het onevenwicht in de redoxbalans in de cad2-1 mutant en het gebrek aan een correcte, kortstondige ethyleensignalering in een suboptimale acclimatie en, bijgevolg, een sterker Cd-effect. Zoals reeds aangetoond is ethyleensignalering belangrijk voor de stimulatie van het GSH-metabolisme in bladeren onder Cd-blootstelling en is ethyleensignalering betrokken bij de Cd-geïnduceerde oxidatieve stress. In deze studie werd er getracht om te verduidelijken hoe ethyleensignalering betrokken is bij de acute Cd-geïnduceerde wortel- en bladresponsen en hoe deze worden beïnvloed door verhinderde ethyleensignalering. Door dezelfde responsen, die hierboven reeds aan bod zijn gekomen, te bestuderen in WT planten en ein2-1 mutanten, stelden we vast dat de rol van ethyleensignalering tijdens de Cd-geïnduceerde stressresponsen grotendeels gelijkaardig is in blad en wortel. Meer bepaald hebben we aangetoond dat in beide organen de timing en de kortstondigheid van de ethyleenrespons beïnvloed wordt onder een verstoorde ethyleensignalering en dat dit resulteert in een vertraagde oxidatieve burst en een gebrek aan GSHstimulatie onder Cd-blootstelling. Deze verstoorde timing van de stressresponsen heeft een negatief effect op stabilisatie. Omdat ACC-biosynthese gestimuleerd wordt onder Cd-blootstelling en dit effect versterkt wordt door GSH-deficiëntie, was ons doel om het belang van ACCbiosynthese tijdens de acute Cd-geïnduceerde stressrespons te verduidelijken. Ditmaal werden de wortel- en bladresponsen in WT A. thaliana planten vergeleken met de responsen in de acs2-1acs6-1 dubbele knockout mutant aangezien de twee ACC-synthase isozymen, ACS2 en ACS6, grotendeels verantwoordelijk zijn voor Cd-geïnduceerde ethyleenbiosynthese. De resultaten toonden aan dat de impact van een verstoorde ACC-productie beperkt bleef tot 24 u na blootstelling en dat dit hoofdzakelijk zichtbaar was als een vertraging in GSH-biosynthese. Daarenboven, vindt de Cd-geïnduceerde oxidatieve burst in WT planten plaats na 24 u blootstelling, terwijl deze respons vertraagd is in de bladeren van de ACCbiosynthese mutant en ontbreekt in de wortels van de mutant. Op basis hiervan kunnen we concluderen dat de Cd-geïnduceerde oxidatieve stress in WT planten beter gecontroleerd wordt in vergelijking met de ACC-biosynthese mutant. Samengevat geeft deze studie een gedetailleerd beeld van het tijdsverloop van de acute Cd-geïnduceerde stressresponsen en -signalering die acclimatie voorafgaan. Op basis van deze data stellen we voor dat de Cd-geïnduceerd GSH-depletie in de wortels nodig is voor het uitlokken van een gepaste stress signaleringsrespons en hebben we aangetoond dat GSH-deficiëntie een vroegtijdige stressrespons veroorzaakt wat resulteert in suboptimale acclimatie. Daarenboven werd aangetoond dat een verstoorde ethyleensignalering en -biosynthese de GSHstimulatie onder Cd-stress verhindert en een vertraging in de oxidatieve burst teweegbrengt. Dit zal op zijn beurt leiden tot een verstoorde signaleringsrespons en een suboptimale of, op zijn minst, vertraagde acclimatie. |
