Abstract: Elastin-Like Proteins (ELP) are highly versatile biomaterials with significant potential in tissue-engineering, due to their design flexibility, mechanical properties and tunable stimuli responsiveness. This study establishes a robust framework for the synthesis of modular ELP constructs featuring intermittent peptide domains. Response Surface Methodology is employed to systematically optimize protein expression, significantly enhancing yield. Analysis of lower critical solution temperature of ELP suggests a correlation between the molecular architecture and resulting physicochemical properties in regards to the incorporated peptides and their location. Furthermore, In vitro biocompatibility analysis confirm high cell viability, validating these modular ELPs as promising candidates for advanced biomedical applications.
