dc.description.abstractTraumatisch ruggenmergletsel (TSCI) veroorzaakt een inflammatoire respons die bijdraagt aan weefselschade. B-cellen lijken hierbij een belangrijke rol te spelen. De MIF/CD74 as is betrokken bij verschillende inflammatoire aandoeningen, maar de rol ervan in B-celgemedieerde pathologie na SCI is nog onduidelijk. Deze studie onderzocht de bijdrage van de MIF/CD74 as aan inflammatoire B-celresponsen na SCI en evalueerde de effecten van inhibitie. Humane primaire B-cellen werden polyclonaal gestimuleerd met de MIF-inhibitor ISO-1 of het anti-CD74-antilichaam Milatuzumab, waarna B-celactivatie en RNA-opbrengst werden bepaald. Daarnaast werden muizensplenocyten behandeld met het CD74-blokkerende construct DRhQ om CD74-oppervlakte-expressie te analyseren. Immunofluorescente kleuringen op ruggenmergweefsel van contusie-SCI muizen (DRhQ of vehicle) werd uitgevoerd om CD74⁺ B-cellen en CD74⁺ microglia/macrofagen in de laesie te detecteren. ISO-1 verminderde CD86-expressie, ERK-fosforylatie en RNA-opbrengst in gestimuleerde B-cellen, wat wijst op inhibitie van inflammatoire signaalroutes. Milatuzumab had geen effect. De validatie van DRhQ was beperkt, aangezien zowel DRhQ als vehicle de CD74-oppervlakte-expressie in splenocyten verlaagden. CD74⁺ B-cellen en CD74⁺ microglia/macrofagen werden aangetoond in het laesiegebied van het ruggenmerg. Deze resultaten ondersteunen een rol voor de MIF/CD74 as in B-celgeassocieerde inflammatie na SCI en vormen een basis voor toekomstige transcriptomische studies naar therapeutische doelwitten.
