dc.description.abstractRadiotherapie is een effectieve kankerbehandeling, maar gaat gepaard met langdurige nadelige effecten op het skelet. Hoewel stralingsgeïnduceerd botverlies en een verhoogd fractuurrisico goed gedocumenteerd zijn, blijven de onderliggende mechanismen onvoldoende begrepen. Eerdere in vitro studies suggereerden dat osteoclasten apoptotische cellen kunnen opruimen via efferocytose, wat mogelijk de bothomeostase beïnvloedt. Daarom onderzocht deze studie of dergelijke interacties ook in vivo gevisualiseerd kunnen worden. Hiervoor werd een imaging-workflow ontwikkeld die tissue clearing en immunofluorescentie combineert om apoptotische cellen en botresidente cellen in het bot van muizen na bestraling te bestuderen. Hoewel tissue clearing succesvol was en apoptotische cellen konden worden gedetecteerd, is verdere optimalisatie van botspecifieke immunokleuringen noodzakelijk. Daarnaast onderzocht deze studie de effecten van mechanische belasting op bestraald bot. Muizen werden blootgesteld aan bestraling en ondergingen unilaterale mechanische belasting van de tibia twee of tien weken na bestraling. De botmicroarchitectuur werd geanalyseerd met micro-computertomografie, terwijl immuunveranderingen werden geëvalueerd via flowcytometrie en orgaangewichtanalyses. Mechanische belasting verbeterde de trabeculaire en corticale botparameters.
