dc.description.abstractHet parodontale ligament (PDL) is een mechanisch veeleisende, sterk geïnnerveerde micro-omgeving. Schwanncellen vereisen continu cholesterol om axonale myelinisatie en zenuwfunctie in stand te houden, hoewel lokale lipidentransportmechanismen slecht begrepen zijn gebleven. Epitheliale celresten van Malassez (ERM) zijn nauw geassocieerd met parodontale zenuwen, wat een cruciale ondersteunende niche suggereert. Deze studie onderzoekt de ruimtelijke koppeling tussen ERM en Schwanncellen en evalueert cholesteroltransportcomponenten in de zich ontwikkelende menselijke tandfollikel (DF) en in volwassen PDL-weefsels. Singleplex- en multiplex immuunhistochemie (IHC) en immuunfluorescentie (IF) op menselijke DF- en PDL-coupes evalueerden celspecifieke markers (TP63, CK14, MPZ, β3-tubuline) en componenten van de cholesteroltransportroute (APOE, LDLR), gevolgd door kwantificering van ruimtelijke afstanden. Weefselanalyse toonde een nauwe ruimtelijke nabijheid tussen ERM en Schwanncellen/zenuwen, die in volwassen PDL significant korter was dan in de zich ontwikkelende DF. Multiplex IHC bevestigde dat TP63-positieve ERM-cellen de kerncomponenten van het cholesteroltransport, APOE en LDLR, tot expressie brengen, waarbij APOE ook aanwezig is in aangrenzende Schwanncellen en zenuwen. Uiteindelijk suggereert dit histologische bewijs van ruimtelijke koppeling en gelokaliseerde transportelementen dat de ERM fungeert als een knooppunt voor lipidenvoorziening, ter ondersteuning van de overleving van Schwanncellen en de parodontale innervatie.
