dc.description.abstractNeuroontwikkelingsstoornissen (NDD's) worden traditioneel bestudeerd in neuronen. Recent bewijs impliceert microglia als bijdragers. Om functies uit te voeren voor de neuroontwikkeling, vertrouwen microglia op een balans tussen de vorming van lamellipodia en filopodia, welke in stand wordt gehouden door actinereorganisatie. Het NDD-risicogen DISC1 verstoort dit evenwicht, wat de microgiale functie aantast. Transcriptoomanalyse van Disc1-locus impairment (LI) microglia identificeerde een neerwaartse regulatie van Diaph2 en Fchsd2, twee actineregulators aan weerszijden van het filopodia-lamellipodia-evenwicht. Hun orthologen in de fruitvlieg, diaphanous (dia) en nervous wreck (nwk), werden onderzocht om te bepalen of deze regulators de morfologie en motiliteit van fagocyten moduleren. RNAi-gemedieerde knockdown onthulde tegengestelde morfologische fenotypen in vitro. Depletie van dia verhoogde de cellulaire circulariteit en oppervlakte, terwijl het filopodia onderdrukte. Knockdown van nwk stimuleerde een filopodia-rijke, asymmetrische morfologie. In vivo migratieanalyse onthulde functioneel verschillende motiliteitsprofielen die consistent zijn met deze architecturen: dia-gedepleteerde hemocyten migreerden sneller maar zonder directionele persistentie, terwijl nwk-gedepleteerde hemocyten trager bewogen met een verhoogde mate van directionele verandering. Deze bevindingen tonen duidelijke rollen aan voor dia en nwk in de actineregulatie van hemocyten, met functionele gevolgen voor de celmotiliteit.
