Abstract: This work is a first attempt to create a protein map of apple leaf material inoculated with Erwinia amylovora. Apple leaf material was inoculated with E. amylovora or mock inoculated. Proteins were extracted from this material and 2-D gel electrophoresis (2-DE) was performed. 2-D gel spots were picked and identified by mass spectrometry. So far, 73 protein spots have been identified, amongst which proteins involved in photosynthesis and in the Calvin-Benson cycle and subunits of ATP-ase. The various 2-D gels were analyzed to identify differentially expressed proteins. Identified proteins upregulated upon E. amylovora inoculation include members of the PR-protein families 2, 5 and 10. PR-2 is the only PR protein shown to be upregulated systemically.
